For what is the Vivaclear mini clarifying filter used?

The Vivaclear mini clarifying filter is used for processing serum, cerebrospinal fluid (CSF) and whole blood. 

Serum and CSF

  •  Transfer 500 μL into an Eppendorf centrifugal filter (0.8 μm) and centrifuge for 5 minutes at 2000 x g.
  • Proceed with 'Wet lab 1: Benzonase'.

Whole Blood

  • Centrifuge whole blood (with heparin) for 10 minutes at 2400 rpm (calculate appropriate g-force).
  • Transfer 500 μL of plasma into an Eppendorf centrifugal filter (0.8 μm) and centrifuge for 5 minutes at 2000 x g.
How to dilute the EDTA?
In the PathoSense reagents package is 1 µM EDTA (1 mL) included. This concentrated solution must be diluted to 10nM (100x dilution) in PCR graded water. 

 

How to store the Zymo Extraction kit?

Store the Zymo Extraction kit at room temperature.

How to make an account on the PathoCloud?

You can make an account by going to cloud.pathosense.com and pressing the register button from the Log In menu. 


Upon your first log in, you will be asked if you want to add a new company, or join an existing company. 

In case you are the first user from your lab, you will need to add a new company with all your lab details. Once this is done, send an email to info@pathosense.com, so we can make sure you have access to all lab functionalities. 

In case the lab is already registered, you will be able to join your lab by pressing ‘Join company’ and entering an invite code that one of your registered colleagues can generate for you in the ‘Companies’ page.

Where to find the 'Join company' code?

You can share an invite code to one of your colleagues by pressing ‘Share Invite’ on the companies page on cloud.pathosense.com

New users can join your lab with this code.

 

How to register a sample in the PathoCloud?

In most cases, the veterinarian will register their own sample through the PathoSense app. In that case, you will see the analysis in your dashboard immediately. 

When you need to register internal samples, or the veterinarian did not register the sample yet, you will need to register it yourself.

One analysis from the same customer can contain multiple samples. 

Log into your account on cloud.pathosense.com. Press ‘New analysis’. In case you are registering it for a known user, select the user from the dropdown. This way the veterinarian can also see the analysis in the app and will  get the result through the app. 

In case the veterinarian is not registered, you can still add their name on the report by checking the ‘Name on report’ checkbox and adding the veterinarian’s name. 

Fill out all known information and enter each sample that is linked to this analysis and submit the analysis. 

Use the “Animal name / number” field for the ID provided by the veterinarian. 

Use the “Internal ID” field for your own lab ID

Use the “Tube label” field if the tube label differs from the internal ID.

How to add the name of the vet to the report if the sample was registered by the lab?

If the veterinarian has a registered account, enter their account as the user for the analysis. This way the veterinarian can also see the analysis in the app and will  get the result through the app. 

 

If the veterinarian is not registered, you can still add their name on the report by checking the ‘Name on report’ checkbox and adding the veterinarian’s name. This way, the veterinarian will not see the analysis and the report will need to be sent to them once the analysis is done.

What is the importance of providing metadata for each sample?

It is very important for a correct analysis of the sample that all sample information is registered correctly. 

The provided host is used to remove host DNA during the data analysis, which speeds up the analysis significantly. 

The provided sample type and anamnesis are used during the annotation of the results. Without this information it is difficult to interpret results and intercept any strange results.

How to add additional info to a sample?

You can add all sample information when registering the analysis, or later edit the analysis by pressing the pencil icon on the right side of the analysis. 

It is very important to fill out all known analysis information. 

Use the “Animal name / number” field for the ID provided by the veterinarian. 

Use the “Internal ID” field for your own lab ID

Use the “Tube label” field if the tube label differs from the internal ID.

How to change the status of the sample?

When a sample arrives at your lab, look up the sample in the dashboard and alter the status from “En route” to “Arrived” by double clicking the status field. Then the veterinarian gets notified through the app that the sample has arrived at your lab. 

The status of the sample will move to “In preparation” when the sample is getting analysed at PathoSense. Once we complete the data analysis, the status moves to “Complete” and the report is sent to your lab email and can be downloaded from the dashboard.
